The main difference that I can see based on my own experience years ago is that the symboled manifest trains (like the CSX "Q" series now) fall under a system-wide coordinated plan and stricter parameters for "scheduling" connections with other trains, interchange cut-offs, blocking, etc. In my day, there was a difference is how crews were paid -- locals were based on hours and went on overtime after 8 hours on duty -- manifest trains were paid according to mileage run, so the sooner you could run off the miles, the more per hour you were making. I don't know how that works today. 100 miles equaled a day's pay, so a run from Flint to GR via Plymouth was 183 miles, or almost "two days" pay.
