There is no doubt at all that toxins, particularly dissolved toxins, pass through a koi's skin. Just consider a pond suffering from raised dissolved copper levels. These levels can be well within what is permitted in drinking water, but high enough to make koi seriously ill or even to kill.
When you have witnessed a pond with this problem (I have seen two) you understand the point all too well.
Pondwater continuously passes through a koi's skin through osmosis, into the bloodstream (admittedly in low levels in a healthy koi). All the dissolved chemicals in the pondwater therefore find their way into the koi's body.
I think I once read that the skin on a doitsu koi was thicker than the skin on a scaled koi. Does anyone have any info on this?
I have no idea about comparisons with human skin. My guess? I would imagine that doitsu skin is much thicker, with more layers and complexity. JR's territory?
